Once its running you can log into the console and update the databases to keep up with any issues you may have, but I have been running PiHole DNS for a good 2 weeks now and have only had to white list 1 domain.
Here is a rather simple guide, i didn't follow it word for word because its rather simple to get going - once you install the OS and SSH into the device you just run one command in the console.
